چکیده

Atopic dermatitis (AD) is a chronic inflammatory skin disease in which patients suffer from red, itching lesions. Current therapeutics target disease-associated immune cells but neglect local skin-specific factors AD and are not suitable for the majority of mild to moderate adult pediatric patients. AHR activation has been described reverse pathology increasingly recognized as new therapeutic target. We use approved drug leflunomide activate aim gather biological evidence repositioning treatment AD. Therefore, primary human keratinocytes were grown into 3D epidermal equivalents stimulated with IL-4 mimic They treated TCDD, (AHR activators) leflunomide’s metabolite teriflunomide (not activating AHR) harvested genome-wide transcriptomic protein expression analysis. find normalize hyperproliferation, restoring barrier, normalizing antimicrobial peptide dampening signaling. Mutations FLG gene strongly affected keratinocyte response IL-4, yet responsiveness. In conclusion, this study shows complete profile agonist dependent under conditions keratinocytes. demonstrate potential through triple action against by proliferation rates, improving barrier function This opens avenues repurposing topical diseases.

The Anti-Inflammatory Drug Leflunomide Is an Agonist of the Aryl Hydrocarbon Receptor

BACKGROUND The aryl hydrocarbon receptor (AhR) is a ligand-activated transcription factor that mediates the toxicity and biological activity of dioxins and related chemicals. The AhR influences a variety of processes involved in cellular growth and differentiation, and recent studies have suggested that the AhR is a potential target for immune-mediated diseases. METHODOLOGY/PRINCIPAL FINDINGS...

Aryl hydrocarbon receptor activation restores filaggrin expression via OVOL1 in atopic dermatitis

Filaggrin (FLG) mutation is a well-confirmed genetic aberration in atopic dermatitis (AD). Genome-wide association studies on AD have revealed other susceptibility genes, for example, Ovo-like 1 (OVOL1). Nonetheless, the relation between FLG and OVOL1 is unclear. Because aryl hydrocarbon receptor (AHR; a ligand-activated transcription factor), plays a role in FLG expression in keratinocytes, we...

A ligand for the aryl hydrocarbon receptor isolated from lung.

The aryl hydrocarbon receptor (AHR) is a ligand-inducible transcription factor that is best known because it mediates the actions of polycyclic and halogenated aromatic hydrocarbon environmental toxicants such as 3-methylcholanthrene and 2,3,7,8-tetrachlorodibenzo-p-dioxin. We report here the successful identification of an endogenous ligand for this receptor; approximately 20 microg was isolat...

Activation of the Aryl Hydrocarbon Receptor Dampens the Severity of Inflammatory Skin Conditions

Environmental stimuli are known to contribute to psoriasis pathogenesis and that of other autoimmune diseases, but the mechanisms are largely unknown. Here we show that the aryl hydrocarbon receptor (AhR), a transcription factor that senses environmental stimuli, modulates pathology in psoriasis. AhR-activating ligands reduced inflammation in the lesional skin of psoriasis patients, whereas AhR...
